Using the Epoch & Take3 Plate
Today’s laboratories are searching for ways to use lower reagent and sample volumes, while increasing throughput and efficiency to keep costs in check.
BioTek’s Take3 plate has a simple but innovative design for measuring multiple low volume samples, which is particularly useful for direct nucleic acid and protein quantification.
The Take3 plate has two uniquely designed fused silica slides mounted by magnets to the housing. The base slide is printed with Teflon to form sixteen 2 mm diameter ‘microspots’. The microspots are the ideal size for pipetting samples as small as 2 microliters. When the lid of the Take3 is closed, the samples are sandwiched between top and bottom slides, held to a nominal 0.5 mm pathlength for each microspot.
The absorbance of the samples in the Take3 plate can be measured using any BioTek microplate spectrophotometer - the fixed pathlengths of the plate allow rapid calculation of concentrations based on a standard 1 cm pathlength.
Pre-programmed protocols in the Gen5 software make it simple to choose the sample type and quickly obtain and calculate data from the Take3 plate.
We’ll use an 8 channel pipettor to place our blanks and DNA samples on the Take3 plate.
Open a Take3 session in Gen5 and select the sample type, either single or double-stranded DNA. We’re using double stranded DNA samples, each with their own buffer blank so all microspots will contain blanks.
After the blanks are read, the Take3 plate is removed and the blanks are wiped off both top and bottom slides with a lab wipe.
Next, pipette the samples onto the microspots. Select all sixteen locations and read the plate. The results are calculated automatically and displayed in an Excel sheet. Raw and calculated data are given as is the blank well information.
You can see that using Take3 makes quantification of one or more 2 µL samples fast, easy and accurate.
